SUMMARY:Statistical Methods for Sequential Categorical Data
DESCRIPTION:Presented by Zuofu Huang\nPh.D. Candidate in Biostatistics\nPh.D. Adviser: Dr. Julian Wolfson \nThe widespread collection of data from mobile and wearable devices has created unprecedented opportunities to study human behavior at fine temporal resolution. One common structure for such data is categorical sequences—ordered\, multinomial observations across many time points. These sequences present unique statistical challenges due to their high dimensionality and complex short- and long-term temporal dependence. The goal of this thesis is to further develop statistical tools motivated by real-world questions on such categorical sequences. We begin by addressing the problem of synthesizing sequential categorical data. The ability to synthesize realistic data in a parametrizable way is valuable for a number of reasons\, including privacy\, missing data imputation\, and evaluating the performance of statistical and computational methods. We propose the paired Markov Chain (paired-MC) method\, a flexible framework that produces sequences that closely mimic real data while providing a straightforward mechanism for modifying characteristics of the synthesized sequences. Next\, we aim to develop and evaluate approaches to identifying “key” sequence positions which distinguish sequence types. We frame this challenge as a regression problem and introduce a variety of regularization techniques that could be applied to achieve interpretable dimension reduction. Finally\, we consider prediction tasks arising in settings such as online prediction\, where only partially observed categorical sequences are available and the goal is to infer behavior in the unobserved portion of the sequence. In this work\, prediction tasks in categorical sequences are framed from a missing-data perspective and addressed through sequence synthesis. This perspective allows both time-point and structural prediction tasks to be handled within a common framework while naturally incorporating uncertainty through repeated synthesis.

SUMMARY:Statistical Methods for Dynamic Prediction of ALS Progression Pathways
DESCRIPTION:Presented by Andres Arguedas Leiva\nPh.D. Candidate in Biostatistics\nPh.D. Advisers: Dr. Mark Fiecas & Dr. Erjia Cui \nLongitudinal data are commonly collected in biomedical studies and used to measure change or progression over time. They can be used for diagnosis\, prognosis\, or even stratification in clinical trials. When the longitudinal data are multivariate\, the modeling complexity is increased due to potential correlations across time within and between variables. Data may also be collected sparsely and irregularly\, decreasing the amount of information available for each subject. One approach to deal with these data is functional data analysis (FDA) methods. These can be used to flexibly model the trajectory over time of these sparsely observed data without strong distributional assumptions. Motivated by a collaboration with the ALS Natural History Consortium\, we propose a series of approaches developed to better understand longitudinal trajectories and their relationship with time-to-event outcomes. First\, we apply a semi-competing risks model to jointly account for death and intermediate disease milestones in ALS progression. Second\, we present a dynamic prediction model that uses sparse multivariate functional PCA to estimate unobserved disease progression trajectories and link them to time-to-death. Finally\, we propose a novel method based on a mixture model of Gaussian Processes to cluster these longitudinal trajectories in a scalable and data-driven way. Together\, the methods developed in this dissertation seek to provide varied approaches to understand longitudinal disease trajectories and connect them to survival outcomes based on sparsely observed data.

DESCRIPTION:The goal of the Aging Work Group (AWG) is to create a collaborative\, supportive community of scholars to facilitate the development of innovative and interdisciplinary aging science. \nThe AWG meets the first Monday of every month. Meetings offer a forum to: \n\n Share new ideas and questions related to scholarship;\nShare draft proposals\, manuscripts or other scholarly products for supportive peer review;\nIdentify new collaborative opportunities; and\nDevelop strategies to advance aging science at the University of Minnesota in cooperation with partner institutions and organizations.\n\nThe Robert L. Kane Endowed Chair in Long-Term Care and Aging provides AWG members with the resources to support: a) external review of extramural grant proposals; and b) scientific editing of grant proposals that emerge in a collaborative fashion from AWG activities. \nIf you are interested in learning more about AWG please explore this site.
